Effects of electroacupuncture at Zusanli and Neiguan points on neuroplasticity-related factors GAP-43 and SYN after ischemic injury in rats

    目的 探讨电针(electroacupuncture,EA)治疗足三里、内关穴对脑缺血大鼠神经可塑性因子GAP-43、SYN 和MAP-2 的影响。 方法 将80 只大鼠按随机数字表法分为假手术组(Sham)组、EA 3 d 组、大脑中动脉阻塞(middle cerebral artery occlusion,MCAO)3 d 组、EA 14 d 组、MCAO 14 d 组,每组16 只。除Sham 组外,其余组采用Longa 线栓法构建MCAO 大鼠模型,EA 组术后每日治疗“足三里”和“内关”穴20 min。平衡木试验评估大鼠运动功能,Western blot 法检测梗死对侧大脑皮层中神经可塑性因子GAP-43、SYN 蛋白的表达,实时荧光定量PCR 法检测GAP-43、SYN mRNA 的表达,免疫荧光法检测GAP-43/ MAP-2、SYN/ MAP-2 的共定位表达。 结果 与Sham 组相比,MCAO 组在各时间点平衡木试验评分显著升高(P<0. 05),MCAO 3 d 组(P<0. 05)和14 d 组(P<0. 05)的GAP-43、SYN mRNA 表达均显著降低,MCAO 14 d 组(P<0. 05)的SYN/ MAP-2 共定位平均光密度表达显著减少,GAP-43和SYN 蛋白表达在MCAO 3 d 组和14 d 组均未见明显差异;与MCAO 组相比,EA 组在1 d、3 d、7 d 时平衡木试验评分未见明显差异,14 d 时平衡木试验评分显著降低(P<0. 05),与MCAO 3 d 组相比,EA 3 d 组GAP-43、SYN mRNA的表达均显著升高(P<0. 05),与MCAO 14 d 组相比,EA 14 d 组的GAP-43、SYN mRNA 表达均显著增加(P<0. 05),以及GAP-43 和SYN 蛋白表达均显著增加(P<0. 05),GAP-43/ MAP-2、SYN/ MAP-2 的共定位平均光密度均显著增加(P<0. 05)。 结论 EA 刺激“足三里”和“内关”穴可改善缺血性损伤后运动功能,其机制可能与促进神经可塑性因子的表达来诱导轴突萌发,进而激活损伤后的神经可塑性有关。

    Abstract:

    Objective To investigate the effect of electroacupuncture(EA) at Zusanli and Neiguan points on neuroplasticity factors GAP-43, SYN and MAP-2 in cerebral ischemic rats. Methods Eighty rats were divided into Sham, EA 3 d, middle cerebral artery occlusion(MCAO)3 d, EA 14 d, and MCAO 14 d groups by a random number table with 16 rats per group. Except for the Sham group, a permanent MCAO rat model was established using the Longa thread embolization method . The EA group was treated at Neiguan and Zusanli points for 20 min daily after surgery. The motor function of rats was assessed by the balance beam test, and protein expression of neuroplasticity factors GAP-43 and SYN in the cerebral cortex contralateral to the infarct was assessed by Western blot. mRNA expression of GAP-43 and SYN was assessed by Real-time quantitative PCR, and colocalized expression of GAP-43/ MAP-2 and SYN/ MAP-2 was determined by immunofluorescence. Results Compared with the Sham group, the MCAO group had significantly higher balance beam test scores at each time point(P<0. 05), GAP-43 and SYN mRNA expression was significantly lower in MCAO 3 d(P<0. 05)and 14 d(P<0. 05) groups, a significantly reduced SYN/ MAP-2 colocalized mean optical density was found in the MCAO 14 d group(P<0. 05), and GAP-43 and SYN protein expression was not significantly different in MCAO 3 d and 14 d groups. Compared with the MCAO group, no significant differences were seen in balance beam test scores at 1, 3, and 7 days in the EA group, and the balance beam test scores were significantly lower at 14 days(P<0. 05). Compared with the MCAO 3 d group, GAP-43 and SYN mRNA expression was significantly increased in the EA 3 d group(P< 0. 05). Compared with the MCAO 14 d group, GAP-43 and SYN mRNA expression was significantly increased(P<0. 05)GAP-43 and SYN protein expression was significantly increased in the EA 14 d group(P<0. 05), and the colocalized mean optical density of GAP-43/ MAP-2 and SYN/ MAP-2 were significantly increased(P<0. 05). Conclusions EA stimulation of Zusanli and Neiguan points improves motor functions after ischemic injury, and the mechanism may be related to promoting expression of neuroplasticity factors to induce axon sprouting and activating neuroplasticity after injury.
