    目的 探讨中药苦参中有效成分氧化苦参碱治疗特应性皮炎(atopic dermatitis, AD)小鼠的药效作用和初步作用机制。 方法 (1)将 KM 小鼠随机分为 5 组:空白对照组、模型对照组、阳性对照组及氧化苦参碱低、高剂量(50、100 mg / kg)组,每日给药 2 次,连续给药 7 次,末次给药后 10 min,于小鼠左后足脚垫内皮下注射右旋糖酐每只 50 μL,观察并记录 15 min 内小鼠舔后足的次数。 (2)将 BALB/ c 小鼠随机分为 6 组:空白对照组、模型对照组、阳性对照组及氧化苦参碱低、中、高剂量(25、50、100 mg / kg)组,除空白对照组外,其余 5 组小鼠均采用 2,4-二硝基氯苯 (DNCB)多次反复刺激皮肤建立 AD 模型,实验第 1 天起各组灌胃给予相应溶媒或药物,连续干预 14 d。 观察并记录 AD 模型小鼠的体重变化,测定脾及胸腺的脏器系数,ELISA 法测定外周血中总 IgE 及 Th2 型细胞因子的含量,HE 染色观察皮损组织病理学变化情况,甲苯胺蓝染色观察皮损组织中肥大细胞的浸润情况。 结果 氧化苦参碱能显著抑制瘙痒模型小鼠的搔抓次数(P<0. 01);灌胃给予氧化苦参碱治疗 14 d 后,AD 模型小鼠湿疹样皮炎缓解;病理切片结果显示,小鼠真皮层增厚减轻、炎性细胞数目减少以及细胞间水肿减轻;真皮层中肥大细胞的浸润数目明显减少,外周血中总 IgE 和 Th 型细胞因子 IL-4、IL-13 的表达显著降低(P<0. 05)。 结论 氧化苦参碱对 AD 模型小鼠有一定的治疗作用,其机制与抑制皮肤真皮层中肥大细胞的浸润、减少 Th2 型细胞因子的表达有关。

    Abstract:

    Objective To investigate the anti-atopic dermatitis (AD) effects of oxymatrine in a murine model of AD. Methods (1) Kunming mice were randomly divided into five groups: Blank Control group(BC), model control group (DEX), positive control group(DXM), OXY-L and OXY-H groups (50 and 100 mg / kg) oxymatrine groups. Vehicle or oxymatrine was orally administered twice daily for 7 times. Ten minutes after the last administration, 50 μL of dextran was injected subcutaneously into the left hind foot pad of each mouse. The number of times the mouse licked the hind foot within 15 min was observed and recorded. (2) BALB/ c mice were randomly divided into 6 groups: blank control group(BC), model control group(DNCB), positive control group (DXM), OXY-L, OXY-M and OXY-H groups ( 25, 50 and 100 mg / kg) oxymatrine groups. Except for the blank control group, the other groups were treated with 2,4-dinitrochlorobenzene (DNCB) solution repeatedly to establish the AD model. From the first day of the experiment, vehicle or drugs were orally administered once daily for 14 d. Changes in weight were observed and recorded, and the organ coefficients of the spleen and thymus were determined. The contents of total IgE and T helper 2 (Th2) cytokines in the peripheral blood were detected using enzyme- linked immunosorbent assay kits. In addition, hematoxylin and eosin staining was used to observe the pathological changes of skin lesions, and toluidine blue staining was used to evaluate the infiltration of mast cells in skin lesions. Results Oral administration of oxymatrine significantly decreased the number of scratches in the itching model mice (P<0. 01). After 14 d of treatment with oxymatrine by gavage, the eczema-like dermatitis of the AD model mice was relieved, and thickening of the dermis, infiltration of inflammatory and mast cells, and intercellular edema were reduced. The expression of total IgE and Th2-type cytokines, such as interleukin IL-4 and IL-13, was significantly lowered in the peripheral blood ( P< 0. 05). Conclusions Traditional Chinese medicine refers to the experience and knowledge summarized by Chinese people in the long-term practice of diagnosis and treatment of diseases. Oxymatrine, an effective component of the commonly used Chinese medicine Sophora flavescens, has a therapeutic effect on AD mice. Its mechanism is related to a reduction in the infiltration of mast cells in the skin dermis and the reduced expression of Th2-type cytokines.
