    目的 探讨分化抑制因子 1 (inhibitor of differentiation 1, Id1)在乙型肝炎病毒复制、肝癌细胞增殖和凋亡中的作用及机制。 方法 使用 RNA 干扰技术将 HepG2. 2. 15 肝癌细胞株的 Id1 基因沉默,作为 siRNA-Id1 组;设计无关序列作为 siRNA-Control 组;使用 Pifithrin-α 抑制 siRNA-Id1 细胞中 p53 的表达,作为 siRNA-Id1 + Pifithrin-α 组;将 HepG2. 2. 15 肝癌细胞作为 Control 组。 依次使用 qRT-PCR、Western blot、ELISA、MTT、流式细胞术 检测乙型肝炎病毒的复制、细胞的增殖和凋亡。 结果 siRNA-Id1 组细胞中 Id1 mRNA 和蛋白的表达量低于 siRNA-Control 组(P<0. 05)。 与 siRNA-Control 组相比较,siRNA-Id1 组细胞中 HBx mRNA 和蛋白的表达量降低(P< 0. 05),上清液中 HBsAg 和 HBeAg 的含量降低(P<0. 05),细胞增殖缓慢(P<0. 05),早期细胞凋亡比例和晚期细胞 凋亡比例升高(P<0. 05),细胞中 Bax 和 Cleaved caspase-3 蛋白的表达量升高(P<0. 05),而 BcL-2 蛋白的表达量降低(P<0. 05)。 Pifithrin-α 可不同程度的抑制 Id1 基因沉默后的上述生物学效应(P<0. 05)。 结论 RNA 干扰 Id1 可以降低乙型肝炎病毒的复制,抑制肝癌细胞的增殖,诱导细胞凋亡,其机制可能与 p53 的过表达有关。

    Abstract:

    Objective To explore the role and mechanism of inhibitor of differentiation 1 (Id1) on replication of the hepatitis B virus, and proliferation and apoptosis of hepatoma cells. Methods The Id1 gene in the HepG2. 2. 15 hepatoma cell line was silenced by RNA interference (siRNA-Id1 group). An unrelated sequence was designed and used for the siRNA-Control group. The expression of p53 in siRNA-Id1 cells was inhibited by pifithrin-α ( siRNA-Id1+Pifithrin-α group) and HepG2. 2. 15 hepatoma cells were used as the control group. The replication of hepatitis B virus, cell proliferation, and apoptosis were detected by qRT-PCR, western blot, ELISA, MTT, and flow cytometry, respectively. Results The mRNA and protein expressions of Id1 in the siRNA-Id1 group were lower than those in the siRNA-Control group (P<0. 001). Compared with the siRNA-control group, the mRNA and protein expressions of HBx in the siRNA-Id1 group were decreased (P<0. 001), the contents of HBsAg and HBeAg in the supernatant were decreased (P<0.001), the proliferation rate of cells was decreased (P<0. 05), the proportions of early apoptosis cells and late apoptosis cells were increased (P<0. 001), the protein expressions of Bax and cleaved caspase-3 in cells were increased (P<0. 001), and the protein expression of BcL-2 was decreased (P<0. 05). Pifithrin-α partly inhibited these biological effects induced by Id1 gene silencing. Conclusions The RNA interference of Id1 reduced the replication of hepatitis B virus, inhibited the proliferation of hepatocellular carcinoma cells, and induced cell apoptosis. The mechanism may be related to the overexpression of p53.
