    目的 通过检测ETFβ在糖尿病肾病中的改变, 探讨其与脂毒性的关系。方法 体内实验采用腹腔注射链脲佐菌素合并单侧肾切除建立糖尿病肾病模型, 并评价肾小管损伤情况, 检测ETFβ在肾皮质中的表达变化。体外建立脂肪酸诱导肾小管细胞NRK 52E凋亡模型, 构建ETFβ重组质粒, 并将其转染至NRK 52E细胞, 观察ETFβ过表达对脂肪酸诱导细胞凋亡的作用。结果 链脲佐菌素合并单侧肾切除诱导的糖尿病肾病大鼠模型中, 肾小管明显损伤, ETFβ mRNA 和蛋白表达均下降。脂肪酸可诱导NRK 52E细胞凋亡, 过表达ETFβ可减少凋亡。结论 糖尿病肾病模型中ETFβ表达下降, 过表达ETFβ可降低脂肪酸诱导的肾小管细胞凋亡。

    Abstract:

    Objective To detect the expression change of ETFβ in diabetic nephropathy rats and study the role of ETFβ in fatty acid-induced apoptosis in renal tubules. Methods Diabetic nephropathy model was established by intraperitoneal injection of streptozotocin and unilateral nephrectomy. In vivo ETFβ expression was detected in renal cortex, as well as tubular injury evaluated. In vitro fatty acid-induced apoptosis in renal tubular cells NRK 52E model was established and ETFβ recombinant plasmid was constructed to be transfected into NRK 52E cells and furtherly to observe the effect of ETFβ over-expression on the fatty acid-induced apoptosis. Results In the rats model of diabetic nephropathy induced by streptozotocin injection and unilateral nephrectomy, ETFβ mRNA and protein expression were decreased as obvious tubular damage occurred. Fatty acids could induce apoptosis in NRK 52E, and ETFβ over-expression reduced the apoptosis. Conclusion The expression of ETFβ is decreased in diabetic nephropathy model, and ETFβ over-expression can reduce apoptosis induced by fatty acid in renal tubular cells.
