昆明小鼠体外受精胚冷冻保存的研究

    在PBS 20?S 0.5mol/L蔗糖中加2.0和3.0mol/L甘油组的胚胎发育率分别为51.4%和44.4%,冷冻效果明显好于其它浓度甘油;在PBS 20?S十2.0mol/L甘油中,加0.5mol/L蔗糖组的胚胎发育率为47.7%,冷冻效果好于其它浓度蔗糖;慢速降温冷冻组的胚胎发育率为48.8%,效果明显好于快速冷冻法;在各期胚胎冷冻后的发育率中,2细胞胚最低,桑椹胚的存活率明显高于4细胞胚和早期囊胚;冷冻保存后体外受精胚的发育率显著低于体内受精胚:112枚经冷冻保存的体外受精胚移植给受体鼠,2/9妊娠。

    Abstract:

    The post-thaw development rates of the embryos frozen in the 2.0 and 3.0mol/L glycerol in PBS 20?S 0.5mol/L sucrose were 51.4% and 44.1%, significantly higher than those of other groups.The post-thaw develop-ment rate of the embryos frozen in the 0.5mol/L sucrose in PBS 20?S 2.0mol/L glycerol was 47.7%,significantly higher than that of other groups. The development rale of the embryos frozen slowly was 48.8%, significantly higher than that of embryos frozen rapidly.The post-thaw development rate of 2-cell embryos was the lowest in four stage embryos. The survival rate of morula was higher than that of 4-cell and blastocyst. The development rate of in vitro fertilized embryos was significantly lower than that of in vivo fertilized ones. A total of 112 frozen-thawed embryos obtained from in vitro fertilization were transferred to recipients, 2/9 of whom became pregnant.
