目的 明确蛹虫草提取物补肾阳的功效及潜在作用机理。 方法 60 只 SD 大鼠分为正常组、模型组、睾丸片组(0. 216 g / kg)、蛹虫草提取物高、中、低剂量组(0. 72、0. 36、0. 18 g / kg),每组 10 只。除正常组外,其余组均采用腺嘌呤(200 mg / kg)灌胃的方法建立肾阳虚大鼠模型,于造模第 15 天起开始给予受试药,连续 28 d。 采用 ELISA 试剂盒检测大鼠血清促性腺激素释放激素(GnRH)、黄体生成素(LH)、睾酮(T)、雌二醇(E2)水平,HE 染色观察肾、睾丸、附睾、精囊腺的组织形态,免疫组化和 Western blot 法检测睾丸 CYP19、CaM、CaMKⅡ表达,RT- qPCR 检测睾丸 CYP19 mRNA 表达。 结果 与正常组相比,模型组大鼠血清 GnRH、LH、T 水平均显著降低(P< 0. 05),肾、睾丸、附睾、精囊腺组织均有明显损伤,睾丸 CYP19 表达降低(P<0. 01)、CaM 和 CaMKⅡ表达显著增加 (P<0. 01),CYP19 mRNA 表达显著降低(P<0. 01);与模型组相比,蛹虫草提取物高剂量组 GnRH、LH、T 含量均升高(P<0. 05),组织损伤缓解,睾丸 CYP19 表达增加(P<0. 05),CaM 和 CaMKⅡ降低(P<0. 05),CYP19 mRNA 水平增加(P<0. 05)。 结论 蛹虫草提取物通过调控 CYP19 / CaM/ CaMKⅡ信号发挥补肾阳功效。 |

Objective To clarify the efficacy and potential mechanism of Cordyceps militaris extract to nourish kidney yang. Methods SD rats were divided into normal, model, testicular tablet (0. 216 g / kg), and Cordyceps militaris extract high, medium, and low dose groups (0. 72, 0. 36 and 0. 18 g / kg, respectively) with 10 per group. Except for the normal group, all other groups were administered adenine (200 mg / kg) by gavage to establish a kidney yang deficiency rat model. Test drug administration was started on day 15 of model establishment for 28 consecutive days. ELISA were used to measure rat serum gonadotropin releasing hormone (GnRH), luteinizing hormone (LH), testosterone (T) and estradiol (E2) levels. HE staining was used to observe the morphology of kidneys, testes, epididymis, and seminal vesicle glands. Immunohistochemistry and Western blot were used to detect CYP19, CaM and CaMKⅡ expression in testes. CYP19 mRNA expression was detected in testes by RT-qPCR. Results Compared with the normal group, serum GnRH, LH and T levels of the model group were significantly reduced (P<0. 05), kidney, testis, epididymis and seminal vesicle gland tissues were all significantly damaged, and CYP19 expression in testes was reduced (P<0. 01). CaMKⅡ expression was increased significantly (P<0. 01). CYP19 mRNA expression was decreased significantly (P<0. 01). Compared with the model group, GnRH, LH and T contents in the high-dose Cordyceps militaris extract group were increased (P< 0. 05), tissue damage was relieved, CYP19 expression in testis was increased (P<0. 05), CaM and CaMKⅡ were decreased (P< 0. 05), and the CYP19 mRNA level was increased (P<0. 05). Conclusions Cordyceps militaris extract exerts a kidney yang effect by regulating CYP19 / CaM/ CaMKⅡ signaling. |
