    目的 探讨红花黄色素( safflower yellow,SY)对链脲佐菌素( streptozotocin,STZ) 诱导的糖尿病肾病( diabetic nephropathy,DN)小鼠的肾保护作用及作用机制。 方法 采用 C57BL / 6J 雄性小鼠,经腹腔注射 STZ( 50 mg / kg) ,连续 5 d 建立糖尿病小鼠模型。 36 只造模成功的小鼠随机分为模型组、SY 低中高剂量组 ( 10 mg / kg、30 mg / kg、90 mg / kg) 。 给药组小鼠分别给予不同剂量的 SY,每日 1 次,连续给药 10 周。 监测糖尿病小鼠的体重、血糖,分析血肌酐( Scr) 、尿素氮( BUN)水平,HE 染色观察肾组织病理形态变化,Western 检测 PKC、P-Raf、Raf、P-ERK、ERK 蛋白的表达水平,ELISA 试剂盒检测 MDA、SOD、GSH 的含量和 TNF-α、IL-6 和 IL-1β 的表达水平。 结果 与正常组比较,模型组的小鼠进食、饮水量增加、体态消瘦、血糖升高。 与模型组相比,给药组小鼠的体重上升、血糖下降、BUN、Scr 的水平降低;肾小球肥大、基底膜增厚和肾小管空泡样病变得到改善;SOD、GSH 含量上升,MDA 含量下降;TNF-α、IL-6 和 IL-1β 的表达下降;PKC、P-Raf、P-ERK 蛋白的表达减少。 结论 红花黄色素可以减弱 DN 炎性损伤和氧化应激损伤,其机制可能与调控 PKC / Ras-Raf- MEK-ERK 通路有关。

    Abstract:

    Objective To study the protective effect of safflower yellow (SY) in mice with diabetic nephropathy (DN) induced by streptozotocin (STZ). Methods DN was induced by the intraperitoneal injection of STZ (50 mg / kg) for 5 consecutive days. Thirty-six male C57 BL/ 6J mice with DN were randomly allocated to a model group, and SY-10 mg / kg, SY-30 mg / kg, and SY-90 mg / kg groups. The three experimental groups were administered SY once daily for 10 weeks. Their blood glucose and body mass were monitored, and blood urea nitrogen ( BUN) and serum creatinine ( Scr) were analyzed. Renal pathology was assessed using hematoxylin and eosin-stained sections. The protein expression levels of PKC, P-Raf, Raf, P-ERK, ERK were measured using Western blot analysis. The contents of GSH, MDA, and SOD, and the expression levels of TNF-α, IL-6, and IL-1β were measured using ELISA kits. Results Compared with a control group, the food and water consumption and blood glucose concentration were higher, and the body mass was lower in the model group. The body mass, blood glucose, BUN, and Scr were higher in all the treatment groups, but the glomerular hypertrophy, basement membrane thickening, and vacuolar degeneration of renal tubules were ameliorated. The contents of SOD and GSH were higher, whereas that of MDA was lower, and the expression levels of proinflammatory factors were lower in the treatment groups. Finally, the protein expression of PKC, P-Raf, and P-ERK was lower in the SY-30 and SY-90 groups. Conclusions SY may ameliorate inflammation and oxidative stress in DN, which might be mediated via the PKC/ Ras-Raf-MEK-ERK signaling pathway.
