    目的 研究 SMN2 外显子 7 在 SMA 小鼠不同组织的列入,筛选与其相关的剪接因子。 方法 通过 RT-PCR 和非变性 PAGE 电泳分析在神经及非神经组织中 SMN2 外显子 7 列入比例,通过 QPCR 筛选分析剪接因子在大脑和脊髓与非神经组织中的 mRNA 水平。 结果 SMN2 外显子 7 在大脑和脊髓组织中列入比例显著高于非神经组织,QPCR 筛选显示 Nova1 / 2 在大脑和脊髓中的表达显著高于大部分非神经组织。通过差异分析显示,Srsf7 / 9 及 Nova2 在 SMA Ⅰ型小鼠脊髓中的表达显著高于对照小鼠。 结论 神经组织中,Srsf7 / 9 和 Nova1 / 2 基因高表达与 SMN2 外显子 7 列入比例显著提高呈正相关,剪接因子 Srsf7 / 9 及 Nova1 / 2 可能参与 SMN2 基因的剪接调控。

    Abstract:

    Objective We examined the inclusion ratio of SMN2 exon 7 in different tissues from SMA mice and screened related splicing factors. Methods Ratios of SMN2 exon 7 inclusion in neural and non-neural tissues were analyzed by RT-PCR and non-denaturing PAGE electrophoresis. The expression levels of splicing factors in the brain, spinal cord, and non-neural tissues were analyzed by QPCR. Results The inclusion ratio of SMN2 exon 7 in brain and spinal cord tissues was significantly higher than that in non-neural tissues. The QPCR screening result showed that the expression of Nova1 / 2 was significantly higher in the brain and spinal cord than in most non-neural tissues. The difference analysis showed that the mRNA levels of Srsf7 / 9 and Nova2 in the spinal cord of SMA type I mice were significantly higher than those in control mice. Conclusions The high expression of Srsf7 / 9 and Nova1 / 2 is positively correlated with significant increases in the inclusion ratio of SMN2 exon 7 in neural tissues, indicating that Srsf7 / 9 and Nova1 / 2 may be involved in the splicing regulation of SMN2.
